Caspase-1及其下游炎症因子在大鼠脊髓损伤中 的表达及意义

Abstract:

目的探讨天冬氨酸特异性半胱氨酸蛋白酶-1(Caspase-1)及其下游炎症因子白介素-1beta(IL-1β)和白介素-18(IL-l8)在大鼠脊髓损伤(spinalcordinjury,SCI)中的表达及意义。方法将成年雄性Sprague-Dawley(SD)大鼠84只随机分为对照组和脊髓损伤组,每组42只。脊髓损伤组应用改良的Allen's重物打击法建立大鼠脊髓损伤模型,对照组只做全椎板切除。以损伤部位为中心取材,HE染色观察脊髓组织病理变化,免疫组化染色检测Caspase-1、IL-1β及IL-18的表达情况。结果对照组大鼠脊髓组织中Caspase-1及其下游炎症因子阳性细胞有少量表达,而脊髓损伤组Caspase-1及其下游炎症因子的阳性表达较对照组显著增高,Caspase-1及IL-18在脊髓损伤后3d表达达到峰值,而IL-1β于损伤后6h表达达到高峰。结论Caspase-1及其下游炎症因子可能参与了SCI后的继发性损伤,并且可能是继发性脊髓损伤的主要损伤因素之一。
