动脉粥样硬化大鼠动脉的基因芯片分析

Abstract:

目的 用基因芯片技术检测正常Wistar大鼠和动脉粥样硬化Wistar大鼠腹主动脉组织中差异表达的基因,以探讨动脉粥样硬化的发病机制。方法 正常对照组大鼠给予正常饮食及生理盐水腹腔注射;动脉粥样硬化模型大鼠给予高脂饮食及维生素D负荷,并以球囊损伤动脉。90d后测定血浆总胆固醇、甘油三酯水平以确定动脉粥样硬化模型建立成功,取正常大鼠和动脉粥样硬化大鼠的腹主动脉进行切片及苏木精-伊红染色观察病理学差异,提取腹主动脉组织mRNA做基因芯片检查。结果 与正常对照组相比,动脉粥样硬化组大鼠血浆内总胆固醇、甘油三脂水平增高(P<0.01);组织切片证实动脉粥样硬化形成;基因芯片检测结果显示511种基因表达上调,1219种基因表达下调,其中部分基因与脂质代谢、炎症反应、氧化应激、细胞凋亡相关。结论 动脉粥样硬化时血管组织多种基因发生表达水平的改变,可能存在复杂的网络调控。对其中重点基因进行深入研究有助于预防动脉粥样硬化发生和改善该病的预后。
