抑制11β-HSD1对滑膜成纤维细胞炎症因子表达的影响

Abstract:

目的观察体外在IL-1β诱导的炎症环境中,滑膜细胞中Ι型11β-羟基类固醇脱氢酶(11β-HSD1)活性在受到干预时,细胞中炎症因子及11β-HSD1表达受糖皮质激素的作用。方法采用SD大鼠原代滑膜细胞为研究对象,以IL-1β作为滑膜细胞炎症诱导因素。实验分为空白组;IL-1β组;IL-1β+皮质酮组;IL-1β+皮质酮+PF915275组;观察各组TNF-α、IL-1β、11β-HSD1表达变化,并分析培养基中活性皮质醇的含量,分析各指标间是否存在的调节关系。结果应用IL-1β诱导后滑膜细胞TNF-α、IL-1β表达显著升高,活性皮质醇转化小与对照无差异,11β-HSD1表达明显高于对照;同时存在皮质酮时,TNF-α、IL-1β表达受到明显抑制,炎症因子所诱发的11β-HSD1的表达升高也受到抑制,明显低于对照组;PF915275能够逆转皮质酮对炎症因子的表达抑制效应,表现为抑制后的TNF-α、IL-1β水平再度升高。11β-HSD1表达再度回升到单纯IL-1β诱导时的水平。结论前炎症因子可刺激滑膜细胞11β-HSD1表达上调,与糖皮质激素共存时可有效增强11β-HSD1转换活性,抑制炎症因子表达,同时下调炎症诱发的11β-HSD1表达升高,这可能是滑膜细胞内存在维持自稳的反馈性调节机制。
