右美托咪啶对大鼠肾缺血/再灌注肾功能的影响

Abstract:

目的探讨右美托咪啶对大鼠肾缺血/再灌注后肾功能及磷酸化STAT3蛋白表达的影响。方法SPF级SD大鼠30只,随机分为缺血再灌注组(I/R组)、假手术组(S组)和右美托咪啶预处理组(Dex组),每组10只。I/R组切除右肾,左肾缺血45min,再灌注60min,造肾I/R模型;Dex组于造模前泵注30minDex;每组于I/R后6h取肾脏制作病理切片,HE染色观察;另取血样,分离血浆,多功能生化仪检测肌酐(Scr)和尿素氮(BUN)含量;ELISA检测肾损伤分子-1(Kim-1)、IL-1β、TNF-α及P-STAT3含量;Western-blot检测肾组织中P-STAT3表达的变化。结果HE染色结果显示,与S组相比,I/R组肾脏损伤严重,炎症明显,而Dex干预组则明显减轻;肾功能及炎性因子检测结果显示,与S组比较,I/R组和Dex组Scr、BUN、Kim-1、IL-1β、TNF-α及P-STAT3含量均显著升高(PP<0.05)。Western-blot检测结果显示,I/R组肾脏中P-STAT3表达增高,Dex可降低P-STAT3表达水平。结论Dex可以减轻由I/R引起的肾功能障碍,抑制炎性反应,降低磷酸化STAT3表达,对肾脏有一定的保护作用。
