Abstract:

用斑马鱼研究了雷帕霉素(RAP)对胚胎的发育、免疫和运动行为的影响.将野生型AB品系胚胎于8—120hpf暴毒于浓度范围为0—5μmol·L-1的RAP溶液中,结果表明RAP在120hpf的半致畸浓度(EC50)为0.024μmol·L-1,主要畸形表型为卵黄囊水肿、卵黄未被吸收和游囊关闭.在运动行为毒性上,自主运动频率(17—26hpf)与对照组相比具显著差异,表现为前期下降而后期上升的模式;接触反应(27—48hpf)也显著不同,表现为低浓度RAP促进而高浓度抑制的模式;低浓度组(0.01、0.05μmol·L-1)的运动能力(120hpf)均显著弱于对照组,高浓度组(0.1μmol·L-1)则无显著性.通过注射罗丹明B标记的异硫氰酸酯右旋糖苷,发现RAP能抑制糖苷吸收,这可能是抑制卵黄吸收并引起胚胎发育畸形的原因之一.借助免疫转基因斑马鱼MPO品系,发现RAP能抑制免疫细胞(嗜中性粒细胞)的荧光表达.
