转基因棉花对根际土壤微生物多样性的影响

Abstract:

本研究应用PCR、T/A克隆、RFLP等分子生物学研究方法分析了两种大田栽培的转基因抗虫棉(SGK321、中棉所41)苗期、花铃期、衰老期根际土壤微生物特异性DNA序列变化,探讨转基因棉花种植对根际土壤微生物区系组成多样性的影响。16SrDNA和5.8SITSPCR扩增产物经T/A克隆分别得到1066、563个阳性克隆,根据酶切图谱进行聚类分析。结果表明,相同生育期的棉花根际土壤微生物区系相似性远高于转基因棉花根际微生物间的相似性。这表明转基因棉花对根际土壤微生物的影响不明显,棉花根际土壤微生物区系差异主要受生育期影响。
