海南地区溶藻弧菌遗传多样性的RAPD 分析

Abstract:

以来源于泰国的菌株TG06003作对照,对采自海南地区的27株溶藻弧菌(Vibrioalginolyticus)环境菌株进行了随机扩增多态性DNA标记(RAPD)分析,其中6株为毒力菌株,另外22株为非毒力菌株。实验共得到70个多态性条带,多态性比率(PPB)为98.6%。基因多样度的算术平均数和加权平均数分别为0.922和0.4544,Shannon’s信息指数的算术平均数和加权平均数分别为0.1391和0.1713。遗传相似度为0.4429~0.8857,遗传距离为0.1214~0.7520。将溶藻弧菌分为毒力菌株和非毒力菌株两个类群进行分析,结果表明,毒力菌株的遗传多样性丰富,它们的基因多样度和Shannon’s信息指数明显高于非毒力菌株;毒力菌株HN08811、HN08155、HN08809、HN08813明显聚为一类,另外2株毒力菌株分别与非毒力菌株HN08304和HN08803聚为一类,溶藻弧菌毒力菌株存在多种遗传型。
