冈村枝管藻叶绿体psbA基因的克隆及序列分析

Abstract:

psbA基因编码光合系统II反应中心的D1蛋白,是叶绿体基因组中一个重要的光调控基因。根据网管藻(Dictyosiphonfoeniculaceus)、髋藻(Myelophycussimplex)、粗粒藻(Asperococcusfistulosus)、索藻(Chordariaflagelliformis)、点叶藻(Punctarialatifolia)、水云(Ectocarpussiliculosus)、铁钉菜(Ishigeokamurae)、绳藻(Colpomeniasinuosa)、网胰藻(Hydroclathrusclathratus)、幅叶藻(Petaloniafascia)等10种藻类的psbA基因高度保守序列,设计引物,利用PCR方法从冈村枝管藻(Cladosiphonokamuranus)基因组DNA中扩增出约750bp的片段,将该片段连接到pMD18T载体上进行序列测定。结果表明:片段长度为737bp,推导的245个氨基酸序列与网管藻、髋藻、粗粒藻、索藻、点叶藻、水云、铁钉菜、绳藻、网胰藻、幅叶藻的D1蛋白相对应的氨基酸序列的同源性均高于96%。该基因序列已被GenBank收录,登录号为EU332142。
