小菊悬浮细胞辐射育种初步研究Ⅰ.基因型、外植体和诱导愈伤组织培养基的选择

Abstract:

以小菊七月红、金莲、七月蜂黄、矮红、盏黄5个品种的叶片为外植体,用MS+20mg?L-12,4D+02mg?L-16BA诱导愈伤组织培养基和MS+20mg?L-16BA+01mg?L-1NAA分化培养基,选出分化能力较强的品种七月红。再以七月红的叶片上部、叶片下部、叶柄、茎段为外植体,在不同培养基(B5、MS)上诱导胚性愈伤组织,发现茎段最易诱导出胚性愈伤组织,培养基以B5添加05mg?L-12,4D+02mg?L-16BA,MS添加10mg?L-12,4D+02mg?L-16BA效果较好;在添加20mg?L-16BA和01mg?L-1NAA的分化培养基(B5、MS)上,茎段再生能力较理想。
