五种菊花近缘植物组织培养研究

Abstract:

以紫花野菊变种、纪伊潮菊、龙脑菊、那贺川野菊和矶菊5个菊花近缘种属植物无菌苗为试材,进行了茎段离体快繁体系和离体叶片不定芽再生体系研究。结果表明最佳茎段增殖培养基分别为紫花野菊变种,MS+1.0mg.L-16-BA+(0.05~0.10)mg.L-1IBA;纪伊潮菊,MS+2.0mg.L-1KT+0.1mg.L-1NAA;龙脑菊,MS+0.5mg.L-1KT+0.1mg.L-1NAA;那贺川野菊,MS+1.0mg.L-1KT+0.1mg.L-1NAA;矶菊,MS+2.0mg.L-16-BA+0.1mg.L-1NAA。紫花野菊变种、纪伊潮菊、龙脑菊、那贺川野菊试管苗适宜生根培养基是1/2MS;矶菊试管苗适宜生根培养基为1/2MS+0.2mg.L-1NAA。5个材料试管苗离体叶片愈伤组织诱导率均较高,但不定芽的诱导率则因基因型而异,仅那贺川野菊和矶菊能诱导出不定芽。那贺川野菊在MS+0.5mg.L-1KT+0.1mg.L-1NAA培养基上不定芽的再生率和增殖系数均最高,分别为75.0%和3.4;而矶菊在MS+1.0mg.L-16-BA+0.5mg.L-1NAA培养基上的不定芽再生率和增殖系数最高,分别达80.0%和5.8。
