受细菌诱导的植物启动子的克隆及其在转基因烟草中的活性

Abstract:

从烟草中克隆了受病原物诱导的植物启动子(pathogeninducibleplantpromoters,PPPs)PPP1、PPP2、PPP3,其长度分别为1308,1170,220bp,序列分析表明它们均含有受细菌诱导的反应元件(Bac)。资料检索表明,PPP3可能是从植物分离的、受细菌诱导的最短的启动子。构建了包括每个PPP启动子和报道基因uidA(编码β葡萄糖醛酸酶,GUS)的转化单元,用它们以及包括来自椰菜花叶病毒35S启动子和uidA单元,在相同条件下分别转化烟草,获得了分别含PPP和35S启动子的4类转基因烟草品系;分子检测表明,转化单元在转基因烟草染色体上能稳定整合。根据对转基因烟草GUS组织化学测定,3个启动子都可以受青枯菌的诱导
