‘砀山酥梨’叶片中Fer2基因的克隆与表达分析

Abstract:

分别以轻度、中度和重度缺铁的‘砀山酥梨’叶片cDNA为模板,正常叶片为对照,运用RACE技术,克隆Fer2基因的cDNA全长为1052bp,编码区828bp,编码276个氨基酸。采用Protparam、InterProScan和ProtFun2.2Server等生物学信息软件分析蛋白质理化性质和结构域特征,结果表明Fer2与‘翠冠’梨叶片的Fer2蛋白和Fer4蛋白等具有较高的相似性,故命名该基因为PbFer2。推测蛋白质相对分子质量为3054.3,理论等电点为5.80。通过Real-timePCR技术,分析PbFer2基因表达量,结果表明轻度缺铁会提高PbFer2的表达量,随着缺铁程度的增加,PbFer2表达量显著下降。
