猪卵巢中脂联素受体与FSHR和CYP19基因表达的发育变化及其相关性研究

Abstract:

为探讨皖南花猪卵巢组织脂联素受体mRNA与雌二醇(E2)合成相关因子mRNA表达的发育性变化及其相关性,用ELISA法测定0、30、45、90和180日龄(n=5)皖南花猪雌性仔猪血液脂联素(Adp)和E2水平,荧光实时定量PCR法检测卵巢组织脂联素受体1、2(adiponectinreceptor1,2;AdpR1,AdpR2)和卵泡刺激素受体(FSHR)、芳香化酶P450(aromataseP450arom)基因CYP19mRNA的表达水平。试验结果显示雌性仔猪血液E2质量浓度、卵巢组织AdpR1、AdpR2和FSHRmRNA表达均有显著或极显著的发育性变化(P<0.05或P<0.01)。血液Adp质量浓度在30d较低,90d较高;血液E2质量浓度在0d最高,30d较低。AdpR1和AdpR2mRNA表达量均表现为30和90d较高,0、45和180d较低;FSHRmRNA表达量在45d达到最高,90d降至最低;CYP19mRNA表达量在0至45d较低,90d时最高。卵巢组织中AdpR1mRNA表达与AdpR2mRNA表达显著正相关(r=0.380,P<0.05);AdpR1mRNA表达与CYP19mRNA表达显著负相关(r=-0.472,P<0.05),与血清E2水平显著负相关(r=-0.416,P<0.05)。结论Adp可能参与对E2合成(或分泌)的调节。
