山豆根多糖及其硫酸酯体外抗DHV-1感染细胞作用的比较

Abstract:

为研制抗1型鸭肝炎病毒(DHV-1)新药成分,用水煎醇沉法提取山豆根多糖(BSRPS),并采用氯磺酸-吡啶法制备其硫酸化产物山豆根多糖硫酸酯(sBSRPS),然后运用体外细胞培养法比较研究BSRPS和sBSRPS抗DHV-1感染鸭胚肝细胞的作用。采用直接荧光免疫法比较分析BSRPS和sBSRPS对DHV-1吸附肝细胞作用的差异,并用实时荧光定量PCR法比较分析了最有效药物浓度的BSRPS和sBSRPS对DHV-1在鸭胚肝细胞上的复制、释放的影响。结果表明BSRPS和sBSRPS都具有较好的体外抗DHV-1的作用,sBSRPS的效果优于BSRPS。sBSRPS能有效抑制DHV-1的体外复制和释放,BSRPS仅能抑制DHV-1的复制。两者对病毒吸附鸭胚肝细胞没有明显的作用。结论sBSRPS可以作为抗DVH的新药主要成分之一。
