叔丁醇-水流动相提高环糊精手性HPLC分离性能的分子模拟与机理研究

Abstract:

探索了以丁醇作为流动相,环糊精作为固定相的新型高效液相色谱(HPLC)体系.采用分子模拟法建立和验证了仲、叔、正丁醇环糊精的相互作用模型,在用褪色光度法验证后,以模型为依据选择叔丁醇作为流动相,成功地在Waters515-717-996液相色谱系统上分离了盐酸克伦特罗、硫酸沙丁胺醇、普萘洛尔、酪氨酸和色氨酸5种对映体.实验结果表明,丁醇流动相可通过和环糊精的竞争性相互作用提高色谱固定相对对映体的识别能力.
