芦荟大黄素复合光动力处理对人乳腺癌细胞抑制增殖和促进凋亡的体外观测

Abstract:

目的探讨芦荟大黄素(aloeemodin,AE)介导的光动力对人乳腺癌MCF-7细胞增殖、凋亡的影响及其作用机制。方法AE介导的光动力(PDT)作用于人乳腺癌细胞株MCF-7,采用CCK-8法检测细胞增殖状态;Hoechst细胞核染色观察细胞凋亡的形态学变化;Anne-V/PI双染流式细胞技术检测细胞的凋亡率;DCFH-DA染色检测活性氧物质(ROS)产生;JC-1染色检测线粒体膜电位变化情况;Westernblot检测Caspase-3、Bcl-2蛋白表达的变化。结果AE介导的光动力作用显著抑制人乳腺癌MCF-7细胞的生长,其抑制作用呈一定的剂量效应关系(P<0.05);AE介导的光动力作用人乳腺癌MCF-7细胞发生凋亡和继发性坏死的比率均显著高于单纯光照射组、单纯AE光敏剂处理组和空白对照组(P<0.05),而后3组间无明显差异(P>0.05);AE介导的光动力在能量密度为12J/cm2(药物浓度10μmol/L,光功率密度40mW/cm2持续照射时间为300s)作用后Hoechst细胞核荧光染色可观察到细胞核固缩、碎裂,可见凋亡小体;DCFH-DA荧光染色发现PDT组细胞内ROS水平升高;JC-1荧光染色发现PDT实验组较对照组细胞膜电位下降;Westernblot检测发现Caspase-3蛋白表达升高,Bcl-2蛋白表达略有降低。结论AE介导的光动力能有效抑制人乳腺癌MCF-7细胞生长并诱导其发生凋亡,且线粒体通路参与了AE介导的光动力诱导人乳腺癌细胞凋亡的过程。
