MACC1在人食管鳞癌中的表达及对侵袭的影响

Abstract:

目的分析结肠癌转移相关基因1(metastasis-associatedincoloncancer-1,MACC1)在人食管鳞癌组织中的表达及其与临床病例特征间的关系;研究MACC1基因在食管鳞癌侵袭转移中的生物学功能。方法采用免疫组化S-P法检测MACC1在食管鳞癌组织中的表达,结合临床病理特征分析MACC1表达与食管鳞癌侵袭、迁移之间的关系及其临床意义;采用脂质体法将质粒shRNA-MACC1转染食管鳞癌细胞株ECA109,采用G418压力培养筛选稳定转染ECA109阳性细胞,通过荧光定量PCR、Westernblot检测ECA109阳性细胞MACC1基因和蛋白的表达,通过MTT、Transwell检测ECA109阳性细胞增殖能力、侵袭能力的变化。结果免疫组化结果显示食管鳞癌组织MACC1阳性率(94.1%)显著高于癌旁组织(37.3%),差异具有统计学意义(P<0.05);食管鳞癌上皮细胞中MACC1强阳性率(90.0%)显著高于非典型增生食管上皮细胞(15.0%),差异具有统计学意义(P<0.05),未明显增生食管上皮细胞中MACC1表达为阴性;在性别、年龄、肿瘤大小、病变部位、分化程度以及浸润深度分组比较中,MACC1蛋白表达差异无统计学意义(P>0.05)。在体外实验中,随着MACC1表达下调,ECA109细胞增殖能力、侵袭迁移能力明显下降。结论MACC1与食管鳞癌侵袭迁移密切相关;MACC1在非典型增生食管上皮和食管鳞癌组织中异常表达,在由异常增生到食管鳞癌的过程中,MACC1蛋白表达随之升高,提示MACC1基因与食管鳞癌发生、发展密切相关。
