稳定过表达Wnt5a对K562细胞生物学性状的影响

Abstract:

目的构建稳定过表达Wnt5a的K562细胞株,并观察稳定过表达Wnt5a对K562细胞生物学性状的影响。方法构建质粒pSEB-Wnt5a;包装得到含有逆转录病毒的上清,感染K562细胞,筛选出感染成功的细胞;Westernblot检测蛋白的表达;细胞计数、瑞氏染色、透射电子显微镜观察对细胞生物学性状的影响。结果成功构建了质粒pSEB-Wnt5a;筛选得到了感染成功的细胞;感染pSEB-Wnt5a逆转录病毒的K562细胞Wnt5a蛋白表达水平明显高于感染空病毒载体的细胞;与K562-vector细胞相比,K562-Wnt5a细胞的增殖被明显抑制,并出现了向成熟分化的形态学特征。结论成功构建了稳定过表达Wnt5a的K562细胞株;稳定过表达Wnt5a抑制K562细胞增殖并可以促进其向成熟分化,提示Wnt5a在白血病发生中可能起到抑癌基因的作用。
