大鼠下丘脑神经元培养的新方法

Abstract:

目的通过对大鼠下丘脑神经元培养方法的改进研究,寻找更佳的下丘脑神经元培养方法。方法基于目前常用的Neurobasal+B27+GlumaxⅠ的神经元培养体系,采用多重过滤替代原来的取上清过滤的方法处理神经元,并对换液方式作出相应调整。以NF-200对细胞进行免疫组织化学染色鉴定。通过观察比较培养下丘脑神经元的细胞密度、神经元轴突长度和胞体面积等指标,对培养方法作出评价。结果2种培养方法均获得良好的培养效果,改进的方法在细胞密度,3、7、10、12d4个时间点均优于原培养方法;神经元轴突长度和胞体面积方面,新方法只在3d时优于原方法,至7d时二者差异已不明显。结论新方法具有稳定的可重复性,可应用于神经元的培养实验。
