静磁场对成骨细胞增殖活性和细胞表面超微结构的影响

Abstract:

目的观察不同强度和作用时间的静磁场对体外培养成骨细胞的增殖活性及其表面超微结构的影响,探索静磁场加载对成骨细胞增殖等功能活性影响的机制。方法将体外培养成骨细胞分别于静磁场加载装置0、8、50、160mT磁场强度下给予24、48、72h的静磁场加载。应用MTT法检测成骨细胞的增殖活性;应用倒置相差显微镜和扫描电镜观察细胞形态及其表面超微结构的变化。结果与不给予静磁场加载对照组比较,培养成骨细胞分别于8、50、160mT静磁场加载24、48、72h后,其增殖活性出现不同程度的增加;其细胞形态、数量及分布上无差异;而随加载时间延长,细胞表面变得凹凸不平,微绒毛增多增粗增大,可见融合成囊泡及大囊泡,且8mT和50mT静磁场的作用明显。结论8、50、160mT静磁场随加载时间延长,体外培养成骨细胞增殖活性出现不同程度增加,细胞表面超微结构出现不同程度改建。
