弹性纤维系统在小鼠支气管肺发育不良中的动态表达规律

Abstract:

目的观察支气管肺发育不良模型小鼠肺发育过程中弹性蛋白及其相关蛋白的表达情况,探讨弹性蛋白平衡紊乱在支气管肺发育不良(bronchopulmonarydysplasia,BPD)中的作用。方法新生1d内的C57BL/6小鼠按随机数字表法分为两组:BPD组及对照组,BPD组应用高浓度氧暴露建立BPD模型,运用HE染色和免疫组化法观察各组小鼠肺组织形态学改变,弹性纤维染色观察各组小鼠弹性纤维含量,荧光定量PCR技术观察弹性蛋白及其相关蛋白mRNA在各组小鼠中随时间变化的表达情况。结果与对照组相比,BPD组小鼠第3天开始肺部次级脊形成受阻,直至21d可见肺泡融合导致正常肺泡结构消失,次级脊消失,弹性纤维和弹性蛋白明显增加且紊乱无序的沉积在肺泡间隔上;BPD组弹性蛋白及其相关蛋白基因的表达显示:Tropoelastin及其相关合成蛋白Fibrillin-1、Fibrillin-2、Fibulin-4、Fibulin-5mRNA的表达在给氧21d时有不同程度的增加(P<0.05),弹性蛋白交联酶LoxmRNA在给氧3~21d较对照组明显增加(P<0.01),弹性蛋白降解酶NeutrophilelastasemRNA在给氧1~14d表达较对照组增加(P<0.05),21d时减少至与对照组无明显差异(P>0.05)。结论弹性蛋白平衡是肺泡发育的重要机制,弹性蛋白平衡紊乱在BPD小鼠病理过程中起着重要作用。
