人端粒酶逆转录酶启动子转录活性的磁共振成像研究

Abstract:

目的探索磁共振成像技术检测人端粒酶逆转录酶(humantelomerasereversetranscriptase,hTERT)启动子靶向转录活性的可行性。方法构建慢病毒Lenti-hTERT-Fth-flag-Puro和对照Lenti-CMV-Fth-flag-Puro载体,将上述载体分别转染端粒酶阳性的肺腺癌(A549)和端粒酶阴性的原代人牙周膜成纤维(humanperiodontalligamentfibroblasts,HPDLFs?)细胞。进行免疫荧光染色和Westernblot检测细胞Fth-flag表达情况,采用普鲁氏蓝铁染色检测细胞摄铁量,MR扫描观察细胞T*2WI信号和T*2值改变。结果A549细胞转染Lenti-hTERT-Fth-flag-Puro载体后,免疫荧光染色和Westernblot检测结果显示转染细胞表达Fth-flag蛋白,未转染细胞不表达Fth-flag蛋白,普鲁氏蓝铁染色示转染细胞摄铁量较未转染细胞显著增加,MR扫描显示转染细胞T*2WI信号(1340.86±49.21)和T*2值(12.10±0.92)较未转染细胞(2049.08±87.58),(21.44±1.15)显著降低(P<0.05);HPDLFs细胞转染Lenti-hTERT-Fth-flag-Puro载体后,细胞无Fth-flag蛋白表达,细胞摄铁量较未转染细胞无变化,T*2WI信号(2225.42±73.43)和T*2值(24.01±1.13)较未转染细胞T*2WI信号(?2148.48±?54.74)和T*2值(23.39±1.72)无显著差异(P>0.05)。而转染对照Lenti-CMV-Fth-flag-Puro慢病毒载体后,A549和HPDLFs细胞均表达Fth-flag蛋白,转染细胞摄铁量较未转染细胞均显著增加,转染细胞T*2WI信号[A549:(1137.38±60.59),HPDLFs:(1299.16±53.42)]和T*2值[A549:(9.61±1.17),HPDLFs:(11.54±0.74)]均比未转染细胞显著下降(P<0.05)。结论MR铁蛋白报告基因成像技术可用于检测hTERT启动子转录活性,构建的慢病毒载体有望为恶性肿瘤的靶向诊断提供一种新的方法。
