高强度聚焦超声治疗小鼠皮下瘤后IL-6和IL-10表达的变化

Abstract:

目的探讨高强度聚焦超声(HIFU)治疗小鼠结直肠癌皮下瘤的有效性和治疗前后IL-6和IL-10表达的变化情况。方法采用结直肠癌细胞系,建立BABL/c小鼠皮下瘤模型,数量60只;采用JC-200肿瘤治疗系统,分为6组:对照组(荷瘤假照组),治疗后1、3、5、7d及14d组,眼球取血,收集肿瘤组织,石蜡包埋后HE染色观察组织病理变化,并检测残余肿瘤组织和血清中IL-6和IL-10的表达水平。结果HIFU能有效地消融肿瘤组织,靶区组织随时间的变化出现以水肿、出血、炎症细胞浸润、坏死、纤维增生为主的病理学变化。HIFU消融后肿瘤组织及外周血IL-6和IL-10的表达均表现为上升后下降的趋势。HIFU后1、3、5、7d血清中IL-6的浓度均显著高于对照组(P<0.05),14d检测,IL-6的表达则明显下降(P>0.05);血清中IL-10的浓度与对照组相比在1、3d明显升高(P>0.05),1d后达到最高峰,而血清IL-10的浓度在5、7、14d组与对照组相比,无明显差异(P>0.05)。结论HIFU能有效治疗结直肠癌,且肿瘤残余组织和血清中IL-6和IL-10的表达早期升高,后期IL-6明显降低,而IL-10无明显变化。
