IGF-1和IGF-1R在无功能性垂体腺瘤中的表达及意义

Abstract:

目的探讨胰岛素样生长因子1(insulin-likegrowthfactor1,IGF-1)和胰岛素样生长因子1型受体(insulin-likegrowthfactor1receptor,IGF-1R)与无功能性垂体腺瘤侵袭性和复发性的关系。方法60例无功能性垂体腺瘤分为侵袭性复发性组(16例)、侵袭性非复发性组(24例)和非侵袭性非复发性组(20例)。采用qRT-PCR检测IGF-1和IGF-1R的mRNA表达水平,采用免疫组织化学法和Westernblot检测IGF-1、IGF-1R和其下游信号分子血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)的蛋白表达水平。结果IGF-1和IGF-1R的mRNA表达水平及IGF-1、IGF-1R和VEGF的蛋白表达水平,侵袭性复发性组高于侵袭性非复发性组(P<0.05),侵袭性非复发性组高于非侵袭性非复发性组(P<0.05)。相关性分析结果显示,在侵袭性复发性、侵袭性非复发性无功能性垂体腺瘤组织中IGF-1和IGF-1R蛋白表达量呈正相关(r=0.85,r=0.80,P<0.05),IGF-1R和VEGF蛋白表达量呈正相关(r=0.74,r=0.70,P<0.05);在非侵袭性非复发性无功能性垂体腺瘤组织中IGF-1和IGF-1R蛋白表达量呈正相关(r=0.73,P<0.05),IGF-1R和VEGF蛋白表达量无明显相关性(P>0.05)。结论IGF-1、IGF-1R和VEGF在侵袭性和复发性无功能性垂体腺瘤中表达上调,三者协调作用,与无功能性垂体腺瘤的侵袭性和复发性恶性行为的发生、发展可能相关。
