重复经颅直流电刺激对阿尔茨海默大鼠学习记忆能力的影响

Abstract:

目的观察重复阳极经颅直流电刺激(transcranialdirectcurrentstimulation,tDCS)对阿尔茨海默病(Alzheimer’sdisease,AD)大鼠学习记忆能力的影响。方法将36只SD大鼠分为假手术组、AD组、20μA刺激组、60μA刺激组、100μA刺激组和200μA刺激组,每组6只。AD组和刺激组的海马注射Aβ1-40制作AD模型。术后2周,刺激组接受重复阳极tDCS,而AD组和假手术组接受假刺激。tDCS后,用Morris水迷宫和Y迷宫检测大鼠学习记忆能力,尼氏染色观察大鼠海马神经元形态变化。结果Morris水迷宫结果显示,60、100、200μA刺激组的潜伏期均较AD组明显降低(P<0.05)。大鼠经过原平台的次数和原平台象限游泳时间各组间差异并没有统计学意义(P>0.05)。Y迷宫结果显示100μA和200μA刺激后,大鼠学习记忆成绩较AD组均有明显改善(P<0.05)。尼氏染色显示tDCS后大鼠海马神经元胞质中尼氏体数量增加,染色加深。结论重复阳极tDCS能改善AD大鼠学习记忆功能障碍。
