Abstract:

目的研究G蛋白偶联受体激酶4(GRK4)对氧化低密度脂蛋白(ox-LDL)诱导的血管平滑肌细胞增殖的影响。方法以大鼠胸主动脉平滑肌细胞株(A10细胞)为靶细胞,检测在不同浓度的ox-LDL(10、20、30、40、60、80mg/L)刺激下A10细胞的增殖情况;观察ox-LDL(40mg/L)刺激下A10细胞GRK4表达变化;用siRNA干扰A10细胞GRK4表达后,检测ox-LDL对A10细胞增殖的影响。Westernblot检测GRK4蛋白表达,CCK-8检测细胞增殖活力。结果用不同浓度ox-LDL刺激A10细胞,发现ox-LDL(40mg/L)刺激下细胞增殖幅度可达78.3%(P<0.05),且细胞GRK4蛋白表达增加[实验组(0.3679±0.0491),对照组(0.1930±0.0385),P<0.05];与对照组相比,用siRNA干扰A10细胞GRK4表达后ox-LDL对A10细胞增殖的影响明显减弱[对照组(1.0501±0.3029),ox-LDL刺激组(1.9291±0.3900),siRNA组(1.4032±0.1644),P<0.05]。结论ox-LDL可增强GRK4表达,干扰细胞GRK4表达后可减弱ox-LDL诱导的大鼠平滑肌细胞的增殖。
