硫化氢对糖尿病大鼠心肌纤维化及NF-κBp65和TNF-α表达的影响

Abstract:

目的探讨气体信号分子硫化氢(H2S)对糖尿病大鼠心肌纤维化以及TNF-α和NF-κBp65表达的影响。方法将30只雄性成年SD大鼠分为3组:对照组(Control组)、糖尿病大鼠组(STZ组)、硫化氢干预糖尿病大鼠组(STZ+H2S组),每组10只。用腹腔注射链脲佐菌素(STZ,40mg/kg)建立糖尿病模型;Control组每天腹腔注射生理盐水;STZ+H2S组每天腹腔注射硫氢化钠(H2S供体)溶液(100μmol/kg),8周后处死大鼠,HE染色观察心肌组织病理学变化;Westernblot检测Ⅳ型胶原蛋白和NF-κBp65和TNF-α的表达。结果与STZ组相比,H2S干预后的心肌细胞间质纤维化明显改善,而与Control组相比,STZ组的Ⅳ型胶原及TNF-α和NF-κBp65蛋白的表达明显增加(P<0.05);与STZ组相比,STZ+H2S组Ⅳ型胶原及TNF-α和NF-κBp65蛋白的表达则明显减少(P<0.05)。结论H2S改善糖尿病大鼠心肌纤维化,抑制NF-κB/TNF-α的信号通路是其可能机制之一。
