Sulfiredoxin-1对大鼠星形胶质细胞缺血损伤的保护作用

Abstract:

目的探讨小分子蛋白Sulfiredoxin-1(Srxn1)对大鼠星形胶质细胞缺血损伤的保护作用以及Srxn1是否参与活化NF-κB炎症信号通路的分子机制。方法通过H2O2处理、氧糖剥夺/复氧(oxygen-glucosedeprivationfollowedbyrecovery,OGD/R)模型,模拟体内脑缺血再灌注损伤;通过慢病毒载体片段Lenti-Srxn1-sh1和Lenti-Srxn1-sh2干扰Srxn1的表达,采用MTT法检测两种损伤模型中细胞的存活率,采用双荧光免疫细胞化学法检测NF-κB入核情况,Westernblot检测p-NFκBp65、MIF的蛋白表达。结果在H2O2和OGD/R模型中,两组慢病毒干扰片段均可以降低Srxn1蛋白的表达。与Control组比较,干扰Srxn1基因能够明显降低细胞存活率,增加NF-κBp65入核,使p-NFκBp65和巨噬细胞游走抑制因子(macrophagemigrationinhibitoryfactor,MIF)蛋白表达显著增加,差异具有统计学意义(P<0.05)。结论Srxn1作为体内重要的神经保护因子,可以减少大鼠星形胶质细胞缺血性损伤的作用机制之一可能是与其激活NF-κB信号通路相关。
