PilotstudyonbindingofbovinesalivaryproteinstogritsilicatesandplantphytolithsDOI: 10.11813/j.issn.0254-5853.2013.E3.E87 Keywords: Bovinesalivaryproteins,Massspectrometry,Phytoliths,Silicates,Toothenamel Abstract: Mostlyfedwithgrassinfreshorconservedform,cattleandotherlivestockhavetocopewithsilicatedefencebodiesfromplants(phytoliths)andenvironmentalsilicates(grit),whichabradetoothenamelandcouldadditionallyinteractwithvarioussalivaryproteins.Todetectpotentialcandidatesforsilicate-bindingproteins,bovinewholesalivawasincubatedwithgrass-derivedphytolithsandsilicates.Interactionsofsalivaryproteinswithpulverizedbovinedentalenamelanddentinewereadditionallyanalysed.Afterintensewashing,thepowderfractionswereloadedonto1D-polyacrylamidegels,mostprominentadhesiveproteinbandswerecutoutandproteinswereidentifiedbymassspectrometrywithinthreeindependentreplicates.Allmaterialsweremainlyboundbybovineodorant-bindingprotein,bovinesalivaryprotein30×103andcarbonicanhydraseVI.Thephytolith/silicatefractionshowedadditionalstrongerinteractionwithhaemoglobinβandlactoperoxidase.Conceivably,thebindingoftheseproteinstothesurfacesmaycontributetobiologicalprocessesoccurringonthem.
