大豆籽粒氨基酸含量的QTL分析

Abstract:

以“冀豆12×黑豆”构建的包含186个家系的F8代重组自交系(RIL)群体为材料,分析16种氨基酸含量,采用性状-标记间单向因素方差分析法(ANOVA)、复合区间作图法(CIM)和完备区间作图法(ICIM)对各氨基酸含量的相关QTL进行检测。结果表明:群体中各性状表型均呈现连续性正态分布并超亲分离。3种方法共检测到谷氨酸、精氨酸、丝氨酸、丙氨酸、苯丙氨酸、异亮氨酸、天冬氨酸和赖氨酸含量相关的14个QTL,分布于B1、D1a、D1b、H、I、J和K连锁群中,贡献率为4.91%~17.02%。其中完备区间作图法检测到的苯丙氨酸含量相关QTLqPhe201贡献率最大为17.02%,增效基因来自于黑豆,并且在该标记区间内利用复合区间作图法同时检测到与天冬氨酸、谷氨酸和赖氨酸含量相关的QTL,分别为qAsp20、qGlu20、qLys20。位于B1连锁群中精氨酸含量相关的QTLqArg11在性状-标记间单向方差分析和ICIM两方法中同时检测到,在ICIM中检测到其贡献率为11.79%,增效基因来自于冀豆12。
