一种改良的大豆DNA提取方法

Abstract:

大豆组织中含有较多的蛋白质、糖类、酚类和脂类物质,要从中提取高质量的DNA比较困难。针对这一情况提出一种改良UREA大豆DNA提取方法(mUREA),该方法用异丙醇沉淀DNA,并配制washingbuffer洗涤DNA沉淀。并以大豆叶片和种子为材料,将mUREA法与CTAB法和SDS法进行对比。结果表明,以大豆叶片为材料时,mUREA法提取的DNA产率最高,质量最好;以大豆种子为材料时,mUREA法提取的DNA产率略低于SDS法,但纯度最高。综合来看,mUREA法是一种高效的大豆基因组DNA提取法,从大豆种子和叶片中提取的DNA浓度和纯度都较高,能满足后续各种分子生物学的要求。
