大豆根瘤菌AHM2B菌株培养条件的筛选与优化

Abstract:

试验对大豆根瘤菌AHM2B菌株在YMA、TY、PA、BSE共4种培养基中的生长情况进行了比较,并通过单因素和正交试验确定最佳培养条件。结果表明:该菌株在BSE培养基中生长较快,最佳碳源为蔗糖、氮源为酵母粉、pH8.0、温度为28℃,接种量为4%。在单因素试验的基础上,采用正交试验对培养条件中的蔗糖、酵母粉、pH和接种量4个因素进行优化,得到最佳培养条件为:蔗糖10g,酵母粉3g,MgSO4.H2O0.2g,K2HPO4?0.5g,NaCl0.1g,CaCl2.H2O0.1g,Rh溶液4mL,豆芽汁1000mL,pH7.0,温度28℃,接种量4%。
