三羟异黄酮对全氟辛酸暴露的小鼠肝脏抗氧化能力的影响

Abstract:

将80只SPF级ICR小鼠按单纯随机抽样方法分为正常对照组(等体积生理盐水)、PFOA低剂量组(1mg.kg-1.d-1)、PFOA中剂量组(5mg.kg-1.d-1)、PFOA高剂量组(25mg.kg-1.d-1)、GEN干预对照组(20mg.kg-1.d-1)、GEN干预PFOA低剂量组(GEN:20mg.kg-1.d-1、PFOA:1mg.kg-1.d-1)、GEN干预PFOA中剂量组(GEN:20mg.kg-1.d-1、PFOA:5mg.kg-1.d-1)、GEN干预PFOA高剂量(GEN:20mg.kg-1.d-1、PFOA:25mg.kg-1.d-1)组,每组10只,采用灌胃方法,通过计算肝脏比体重系数,检测肝组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH?Px)、乳酸脱氢酶(LDH)、丙二醛(MDA)及一氧化氮(NO)指标的水平,探讨了三羟异黄酮(GEN)对不同剂量的全氟辛酸(PFOA)致小鼠肝脏氧化损伤的保护作用。结果表明:PFOA导致肝脏肿大,增加了小鼠肝脏系数,并随着剂量增加而加重。高剂量的PFOA能明显增高肝脏组织中MDA及NO含量(P<0.05),低、中、高剂量的PFOA均能提高LDH活性和降低SOD、GSH-Px的活性(P<0.05),而GEN在干预PFOA各剂量组中能显著提高GSH-Px的活性和降低LDH活性(P<0.05),在干预PFOA高剂量组中能提高SOD的活性(P<0.05)。GEN对PFOA致肝脏组织的脂质过氧化损伤有一定的拮抗作用。
