PossibleRoleofCarboxylandImidazoleGroupsintheCatalysisofPummeloLimonoidGlucosyltransferase, PP. 1445-1451 Keywords: chemicalmodification,catalyticresidue,carboxylgroup,imidazolegroup,uridinediphosphateglucosyltransferase,pummeloalbedo Abstract: ?Limonoidbitternessisaseriousprobleminthecitrusindustryworldwide.Limonoidglucosyltransferaseisanenzymethatcatalyzestheconversionofbitterlimonoidintonon-bitterlimonoidglucosidewhileretainingthehealthbenefitoflimonoidsinthejuice.Theimmobilizationofthisenzymeinacolumncansolvethejuicebitternessproblem.Moreinformationaboutthecatalyticresiduesoftheen-zymeisneededinthisimmobilizationprocess.Glutamate/aspartate,histidine,lysine,tryptophan,serine,andcysteineresidueswerechemi-callymodifiedtoinvestigatetheirrolesinthecatalyticfunctionoflimonoidglucosyltransferase.Inactivationoftheenzymefollowingmodi-ficationofcarboxylandimidazolemoietieswasaconsequenceofalossinsubstratebindingandcatalysisintheglucosyltransferreaction.ThemodificationofasinglehistidineresiduecompletelydestroyedtheabilityoflimonoidglucosyltransferasetotransfertheD-glucopyranosylunit.Tryptophanseemedtohavesomeroleinmaintainingtheactiveconformationofthecatalyticsite.Lysinealsoseemedtohavesomedirectorindirectroleinthiscatalysisbutthemodificationofserineandcysteinedidnothaveanyeffectoncatalysis.There-fore,weconcludethatthecarboxylandimidazolegroupscontainaminoacidsareresponsibleforthecatalyticactionoftheenzyme.
