黄芪多糖改善老年大鼠的学习记忆水平及其机制研究

Abstract:

该研究通过观察黄芪多糖对老年大鼠学习记忆的影响,试探究其改善学习记忆能力的机制。实验以自然衰老雌性SD大鼠为动物模型,随机分为老年对照组、老年黄芪多糖低、高剂量组(50,150mg·kg-1)和老年脑复康组(阳性对照,560mg·kg-1),各组大鼠连续灌胃给药60d,另设青年对照组。采用旷场和Morris水迷宫测试大鼠的学习记忆能力,蛋白免疫印迹法观察海马组织相关神经可塑性蛋白N-甲基-D-天门冬氨酸受体(NMDA受体)、钙/钙调蛋白依赖的蛋白激酶Ⅱ(CaMKⅡ)、蛋白激酶A(PKA)、cAMP反应元件结合蛋白(CREB)的磷酸化水平及脑源性神经营养调控因子(BDNF)的蛋白表达水平。研究发现与青年对照组比较,老年大鼠学习和记忆能力均显著下降(P<0.01),海马神经可塑性蛋白表达明显降低(P<0.01);与老年对照组比较,黄芪多糖能明显改善老年大鼠的学习记忆损伤,上调老年大鼠海马区神经可塑性相关蛋白表达水平。结果表明黄芪多糖可能通过改善神经可塑性相关蛋白水平的异常表达,进而改善老年大鼠学习记忆能力。
