燕窝酶解产物中多肽的液相色谱质谱分析

Abstract:

目的:针对燕窝中蛋白质糖基化程度不均一和相对分子质量范围不稳定,以及由此导致的蛋白质难以分离、分析和鉴别的问题,研究1种燕窝中蛋白质和多肽的分析方法。方法:利用分步法从燕窝中提取了蛋白质组分,采用糖苷酶和蛋白酶对蛋白质组分进行了处理,采用生物质谱技术对蛋白质降解物进行了分析,并对其中的多肽进行了序列分析。结果:燕窝中的蛋白提取率为79.7%,采用多级质谱技术从提取蛋白质的糖苷酶和胰蛋白酶处理后的混合物中检测出20种多肽。结论:这些多肽的序列与已有蛋白质数据库中不同种类蛋白的局部序列相似,为富含糖蛋白的动物药成分研究提供了技术参考。
