用多项式分布模型评估低质量DNA模板微卫星分型的可靠性

Abstract:

在使用痕量样品(如博物馆样品、古代样品、法医样品以及非损伤性样品)的分子遗传学研究中,由于DNA质量很低,容易导致扩增错误的产生,包括等位基因丢失和假等位基因.本文建立了1个区别对待纯合子和杂合子的多项式分布数学模型,使用2个参数衡量样品的质量,在此基础上计算多管分型法的分型结果正确率.为了检验此模型,本研究从26只秦岭地区川金丝猴(Rhinopithecusroxellana)个体采集非损伤性毛发样品,使用5个四核苷酸重复的微卫星位点进行扩增.根据此模型,实验获得3次阳性PCR结果时,分型结果正确率能够到达99%.对于样品质量很低、需要过多重复实验的样品,本文还推荐了1种替代的分步分型方法,能够进一步减少重复次数.本模型有助于优化实验条件,得到可靠的分型结果.
