血清抑制高磷介导的磷酸钙盐生成、聚集及其相转变机制

Abstract:

通过向改良杜氏伊格尔培养基(DMEM)加入不同浓度的无机磷酸根离子,对比加入胎牛血清和不加入胎牛血清两种条件,对磷酸钙在生物介质中的生成过程进行了研究.通过上清液中钙离子浓度变化、粒度和Zeta电势的检测以及扫描电子显微镜、透射电子显微镜、红外光谱和十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)观测,发现在没有血清的体系中,外加的磷酸根离子会促进磷酸钙盐的生成和聚集并且促使矿物相以羟基磷灰石的形式存在.然而在有血清的体系中,血清蛋白通过降低溶液的过饱和度,使Zeta电势变得更负并且诱导生成短程有序的弯曲晶格,从而起到抑制磷酸钙盐的生成、聚集及无定形相转变的作用.
