人类β珠蛋白基因HBB及其突变体的mRNA-蛋白质二级结构分析

Abstract:

SNPs能改变基因表达,导致编码氨基酸突变,影响蛋白质功能,这是目前被密切关注的问题.对此,普遍认为蛋白质结构的变化源自氨基酸取代,很少有人注意mRNA结构变化的影响.选取人类β珠蛋白基因HBB及其4个突变体样本,用动态延伸折叠方法构建mRNA二级结构,从有关数据库获得它们的表达产物的二级结构.分别对成熟mRNA和蛋白质做突变前后的结构对比,并进行mRNA-蛋白质相应结构变化的对照.结果发现,蛋白质中规则构象一般为mRNA中的稳定区域编码,而不规则构象(β/γ转角、卷曲)为mRNA中的不稳定区域编码.不稳定区域内的碱基突变引起mRNA二级结构的变化,将会在蛋白质结构中留下“变化印迹”.这种mRNA二级结构和编码蛋白质二级结构的相关性可能作为探寻蛋白质功能变化的一种谋略.
