福建山樱花愈伤组织的诱导及植株再生

Abstract:

为了建立福建山樱花植株再生体系,本文分别以福建山樱花外植体茎段和由茎段诱导出的腋芽的小叶为材料,探讨不同植物生长调节剂种类和浓度配比对福建山樱花愈伤组织诱导及植株再生的影响。结果表明:高浓度的2,4-D有利于福建山樱花愈伤组织诱导,茎段和小叶分别接种于MS+2,4-D2.0mg·L-1和MS+2,4-D2.0mg·L-1+BA0.5mg·L-1,相应的愈伤组织诱导率为93.75%和91.30%。小叶诱导出的愈伤组织接种于MS+2,4-D0.5mg·L-1+BA2.0mg·L-1,不定芽分化率为60.7%。将不定芽转接入1/2MS+BA0.5mg·L-1+IBA0.1mg·L-1进行伸长培养,并在1/2MS+IBA0.6mg·L-1培养基中生根形成完整植株。此外,福建山樱花小叶愈伤组织接种于MS+2,4-D0.1~0.5mg·L-1+BA1.0mg·L-1,还可诱导出体细胞胚。
