大白菜热激转录因子基因家族鉴定及表达分析

Abstract:

本文以大白菜基因组数据为基础,利用生物信息学方法对大白菜热激转录因子(heatshocktranscriptionfactors,HSFs)进行了序列特征分析。结果表明,大白菜至少含有30个HSF基因成员,蛋白质序列长度在239~487氨基酸之间;序列多重比对发现,大白菜HSF基因具有高度保守的DBD结构域和广泛的保守基序;染色体定位揭示了除2个基因分布在Scaffold上,其余28个基因在大白菜的10条染色体上呈不均匀分布,Chr03上分布最多;此外,大白菜和拟南芥HSF基因的系统发育关系比较分析表明,这些HSF基因家族成员可以分为A、B、C三类,A类和B类可进一步分为8个和4个亚组,并且存在8对直系同源基因和4对旁系同源基因,表明HSF基因家族存在于芸薹属和拟南芥属分化之前。RT-qPCR分析表明,高温胁迫显著了诱导了大部分大白菜HSF转录因子基因的表达,表明这些基因广泛地参与了大白菜苗期叶片对高温热激反应。
