短时间低浓度紫杉醇处理能够诱导非中心体依赖性的多极纺锤体组装和非整倍体细胞的产生

Abstract:

紫杉醇是一种能够与微管相结合的药物,被广泛应用于肿瘤的治疗.本研究发现,应用短时间低浓度的紫杉醇处理细胞,能诱导有丝分裂期细胞发生非中心体依赖性的多极纺锤体组装.将细胞内的TPX2去除或在细胞内过量表达Aurora-A激酶,能够减少紫杉醇诱导的多极纺锤体的产生.通过缩时电影活细胞示踪观察发现,经紫杉醇短时间处理的部分细胞可以完成多极细胞分裂,产生多个子细胞;其中部分子代细胞还可以再进行至少一个周期,产生新的子细胞.通过软琼脂板长时间培养实验显示,紫杉醇处理后存活下来的细胞可以形成具有不同于母代细胞染色体数目的子代细胞克隆.由以上结果可知,短时间低浓度紫杉醇处理能诱导非整倍体子细胞的产生.
