高表达蛋白酪氨酸磷酸酶-PEST促进HepG2肝癌细胞体外增殖

Abstract:

为深入探讨蛋白酪氨酸磷酸酶-PEST在肝癌中的作用,构建了真核重组表达质粒pcDNA3.1/PTP-PEST并转染HepG2肝癌细胞,经荧光定量RT-PCR和Westernblot对转染细胞进行筛选,选出稳定高表达PTP-PEST基因的细胞株,并采用MTT及Westernblot对高表达PTP-PEST基因的细胞株的增殖速度和生长因子受体结合蛋白2表达量进行检测,观察到高表达PTP-PEST的细胞株内Grb2蛋白表达上调,细胞增殖速度加快.本研究表明,在HepG2肝癌细胞株内,PTP-PEST可能通过影响Grb2蛋白的表达来促进细胞增殖.
