转基因高表达卵泡抑素1对斑马鱼肌肉生长促进作用研究

Abstract:

抑肌素是转化生长因子?超家族成员,其主要功能是对肌肉的生长发育起负调控作用.在哺乳动物中,抑肌素信号是通过其特异性的与激活素受体ⅡB亚基结合而发挥作用.因此,其信号传递作用可以被其结合蛋白以及ActRⅡB的结合蛋白卵泡抑素所抑制.在小鼠肌肉组织中,高表达卵泡抑素可以导致肌肉组织肌纤维的增生和肥大.为了研究鱼类卵泡抑素是否具有类似的作用,用斑马鱼骨骼肌特异性表达的mylz2启动子,启动斑马鱼卵泡抑素在斑马鱼肌肉组织中的表达.结果发现,多个独立转基因系的肌肉细胞中,成肌调控关键基因myoD和pax7表达升高,且成年斑马鱼体重比对照组略微增加.组织学观察表明,在高表达卵泡抑素的转基因鱼骨骼肌中,肌纤维的数目明显增加,单位面积中肌纤维的数量增加,但肌纤维大小变化不明显.结果表明,高表达卵泡抑素能够通过促进肌纤维增殖的方式促进斑马鱼肌肉生长.
