姜黄素与模拟生物膜相互作用的电化学研究

Abstract:

姜黄素是姜黄根茎提取物中的主要活性成分,具有广泛的抗氧化、抗癌、抗菌、抗炎症等特性.有研究表明,姜黄素能够在一个大致相似的浓度,作用于多种结构上互不相关、横跨多个信号通路的膜蛋白,并且推测姜黄素可能是通过对细胞膜产生直接作用,从而间接影响细胞膜蛋白.本文通过在金电极表面共价连接1,2-二棕榈酰-sn-甘油磷脂酰硫代乙醇(DPPTE)/1,2-二棕榈酰-sn-甘油磷脂酰胆碱(DPPC)磷脂双层膜来构建模拟生物膜,运用电化学交流阻抗和循环伏安等技术手段,研究不同浓度的姜黄素与模拟生物膜的相互作用.结果表明,姜黄素对模拟生物膜的影响呈浓度依赖关系:在较低浓度时,姜黄素能够嵌入外层膜中,改变膜的结构使膜变薄;在较高浓度时,姜黄素能够以寡聚体的形式贯穿双层膜,引起膜阻抗的大幅减小.本研究有助于进一步理解姜黄素的生理活性及其对细胞膜及膜蛋白可能产生的影响.
