A Study of the DNA-Binding Properties of Peptides Containing a Specified Motif by High Performance Affinity Chromatography(HPAC)

Abstract:
n contrast to many sequence-specific DNA binding proteins,for which DNA binding motifs such as thehelix-turn-helix,zinc-finger and leucinezipper have been well understood,a quite new mode of DNA bindingis identified as the S(T)PXX motif After proposing the role of a SPXXXGXP motif of peptides in DNA bind-ing process,we show here that the motif specified above prefers to recognize and bind to DNA with muchlower sequence specificity,using DNA cellulose AC. The novel results show that HPAc method is valuablefor producing accurate,highly reproducible features in elucidating the structural and molecular mechanisms ofnon-specific protein-DNA interaction.
