Genetic analysis of the marine mussel Perna viridis (Bivalvia: Mytilidae) based on mitochondrial COI sequence and its phylogenetic relationship with other congeneric species

Abstract:
Partial mitochondrial COI gene sequences from the gonads (male and female) and somatic tissue of mussel Perna viridis were separately amplified by PCR using the universal primers COIL 1490/COIH 2198. A 661 bp nucleotide sequence of COI gene was obtained. It was found that this gene sequence was identical among different tissues of male individual, and low divergence (0.31%) of COI sequence was detected between male and female of the gonad tissues. Only one haplotype was found in both female and male for the sequence. No doubly uniparental inheritance (DUI) was detected. Based on the COI sequences obtained from this study and those of other Perna species from GenBank, NJ tree, MP tree and Bayesian inference (BI) tree were constructed with PAUP and MrBayes respectively, and phylogenetic relationships among them were analyzed accordingly.
