CO-EXPRESSION OF GLAA AND XYNB OF ASPERGILLUS NIGER IN INDUSTRIAL YEAST

Abstract:
A YIp type plasmid NEW4 was constructed, comprised two different cassettes containing glucoamylase (glaA) gene under the control of the PGK promoter and terminater, and xynlase (xynB) gene controlled by ADH1 promoter and terminater. It was transformed into an industrial strain Sacchromyces cerevisiae 2.346 by co-transformation of a YIp type plasmid BEJ16 carrying G418 resistance so that glaA and xynB were integrated into the chromosome of the strain. The stable engineered yeast expressing extracellular glucoamylase and xynlase was obtained .
