Use of ITS and ISSR markers to identify cultivated strains for Lentinula edodes

Abstract:
16 samples were selected including 12 cultivated strains with similar or confused names for Lentinula edodes, 3 strains for Lentinus tigrinus, and one strain for Lentinus lepideus in this study. These samples were firstly analyzed at the level of species by ITS technique, and then were analyzed by ISSR technique. The sizes of the fragments from 3 species amplified by ITS technique were different, which can be used to identify strains from different species. Two fragments owned to Lentinula edodes strains were obtained and 5 pairs of strains for Lentinula edodes showed the same band patterns each other while strains from different species had different band patterns by ISSR analysis. The results were proved by RAPD analysis. In a conclusion, ITS and ISSR techniques can be used to identify cultivated strains for Lentinula edodes.
