CHROMOSOME NUMERATION AND BEHAVIOR OFNUCLEI DURING BASIDIOSPORE FORMATIONIN PLEUROTUS OSTREATUS

Abstract:
Cytological studies of chromosome numbers and basidiospore formation in Pleurotus ostreatus with light microscope were made at every three hour intervals from the completion of lamella differentiation to mature stage of fruiting body, by using propionic iron haematoxylin and Giemsa staining methods. It was found that some young binucleate basidia were produced during the course of fruiting body development. It took about 18 hours for basidia to develop from karyogamy to completion of meiosis. We found that chromosome number of P. ostreatus is 9 (n = 9) after careful observation at various stages of meiosis, especially at diakinesis. Four nuclei immigrated into four basidiospores respectively, leaving the basidia empty. Most of the basidia at tetrad stage were tetranucieate and a few were quinquenucleate.
